This can be one of the most fun parts of making a manga, or one of the worst. Distinctive, original characters are what will make your world go 'round.
I don't think there is any one given process on how to create characters, it's another one that everyone has their own way of doing.
For me, sometimes a character, complete with personality, just pops out of nowhere and takes up residence next to all the other people living in my head. Sometimes an appearance without a personality shows up (like a shell, pretty and all but nothing inside) or a dis-enbodied personality flits in. In those cases they just chill until their 'other half' decides to come over.
Most of the initial character designs I do are just a simple sketch- usually just a head, sometimes a rough body. Generally these are done so they will leave me the heck alone so I can get back to the stuff I really need to be doing.

When you go to really design your character, keep one thing in mind- you are going to be drawing them over and over and over again, hundreds and maybe even thousands of times.
Don't design something you can't do that with.
Stick with easy things for a bit, until you get the hang of what you're doing. Then move to something more elaborate; as you become more familiar with the design, it will get easier to draw.
On Character Sheets.
A character sheet is generally a set of drawings that shows all the details of one of your characters. It will generally have front, back, side, and 3/4 views as well as mugshots, portraits, etc...

Once the appearance is down, the next thing to figure out is their personality. This will play a big part in what they will and will not do, and pretty musch how they will behave once they are put into your little world.
For example, if someone were to jump out of an alley wearing a monster mask and yell BOO, Joe might scream and faint, wheras Jane would beat the snot out of them.
